What is GHRP-6?

GHRP-6, also known as growth hormone-releasing peptide 6 or growth hormone-releasing hexapeptide, is a synthetic mimetic of ghrelin. Initially discovered as a growth hormone secretagogue receptor (GHS-R) binder, researchers later identified ghrelin as the endogenous ligand for GHS-R. Consequently, GHRP-6 is considered a mimetic of ghrelin. Ghrelin, consisting of 28 amino acids, is involved in hunger signaling, regulating food intake, and stimulating growth hormone (GH) release.

GHRP-6 differs from growth hormone-releasing hormone (GHRH) in amino acid sequence and receptor binding. However, when administered together, GHRP-6 and GHRH exhibit synergistic effects, enhancing GH release. This is attributed to GHRP-6’s impact on both the pituitary gland and hypothalamus.

Comprising only 6 amino acids, including unnatural D-amino acids, GHRP-6 has a molecular weight of 929.48 g/mol. The sequence is as follows: L-Histidine, D-Tryptophan, L-Alanine, L-Tryptophan, D-Phenylalanine, and L-Lysine. The inclusion of D-amino acids in the synthetic peptide allows it to stimulate GH secretion from the anterior pituitary gland while inhibiting the function of somatostatin, a hormone that blocks GH release. GH, in turn, stimulates insulin-like growth factors (IGFs), which play crucial roles in metabolism, cell proliferation, and protein synthesis.

Studies in fish have shown that supplementing feed with GHRP-6 at doses of 100 µg/kg or 500 µg/kg improves growth performance. The lower dose enhances aerobic metabolism, while the higher dose increases GH levels comparable to ghrelin. Furthermore, GHRP-6 has demonstrated neuroprotective effects in ischemic stroke cases. In a gerbil model of stroke, combining GHRP-6 with recombinant epidermal growth factor (rhEGF) improved neurological outcomes and survival.
