What is IGF-1 LR3 ?
IGF-1 LR3 is a synthetic peptide designed to resemble human insulin-like growth factor 1 with improved bioavailability. It is a longer peptide (LR3) that incorporates a substitution of glutamic acid with arginine at the third position. Additionally, an extra 13 amino acids are added to the N-terminal, resulting in a total of 83 amino acids, whereas regular IGF-1 consists of 70 amino acids. These modifications contribute to an extended half-life of IGF-1 LR3, ranging from 20 to 30 hours, compared to 12 to 15 hours for IGF-1. Furthermore, IGF-1 LR3 exhibits three times the potency of IGF-1 by reducing the binding of insulin-like growth factor binding proteins, thereby enhancing its overall biological activity.
IGF-1, similar to insulin, binds to its receptor IGFR1, stimulating cell growth, as well as muscle fiber growth and differentiation. IGF-1 LR3 serves as an efficient alternative to IGF-1 in both in vitro and in vivo studies due to its functional mimicry and effective receptor binding. For instance, research conducted on fetal myocyte proliferation and heart mass in a sheep model demonstrated that IGF-1 LR3 promotes cardiomyocyte proliferation and increases blood flow to the heart. In another study, C2C12 cells were treated in vitro and mice in a tumor model were treated in vivo with IGF-1 LR3, revealing its role in preventing muscle wasting in cancer. By activating the PI3K/AKT pathway and inhibiting the upregulation of muscle wasting factors, IGF-1 LR3 promotes protein synthesis.
