What is Melanotan II?
MELANOTAN II 10mg is a synthetic analog of alpha-melanocyte-stimulating hormone (α-MSH). Acting as an agonist at the melanocortin-3 and 4 receptors (MC3R and MC4R), Melanotan II shares similarities with the prohormone pro-opiomelanocortin (POMC) cleavage peptides, including adrenocorticotropin hormone (ACTH) and α-, β-, and γ-MSH. The melanocortin system, encompassing the central nervous system, plays a role in regulating various physiological functions such as cardiovascular function, energy balance, hunger, glucose and lipid homeostasis, sexual function, pituitary gland functions, emotional behavior, cachexia, drug abuse behavior, pain perception, and anxiety behavior.
In a laboratory study utilizing an MC4R knockout model in rats, increased food intake, body weight gain, and fat accumulation were observed. Additionally, brain administration of melanotan II did not alter feeding behavior, confirming the loss of MC4R function. Subcutaneous treatment with melanotan II in neonate prairie voles resulted in reduced play-fighting in male voles during adulthood and enhanced partner preference in female voles during adulthood.
It has been noted that melanocortin signaling is essential for the antidiabetic effects of leptin. In an experiment, the glucose-lowering effect of leptin was countered by pretreatment with MC3R or MC4R antagonist via intra-cerebroventricular (ICV) administration, highlighting the significance of normal melanocortin signaling in mediating the effects of leptin. However, ICV infusion of MC3R or MC4R agonists alone does not effectively control hyperglycemia in diabetic rats, indicating that increased melanocortin activity alone cannot fully account for the glucose-lowering effect of leptin. Given its wide range of effects, including body weight, metabolism, and thermoregulation, melanotan II serves as a valuable reagent in in vitro laboratory settings for conducting diverse experiments.
