RAD-140, also called testolone, is a nonsteroidal selective androgen receptor modulator (SARM): CAS 1182367-47-0, C20H16ClN5O2, molecular weight 393.8. Each bottle holds 90 capsules at 10mg per capsule.
Background
Radius Health published RAD-140’s design and preclinical profile in 2011 (Miller et al., ACS Med Chem Lett, PMID 24900290). In castrated rats it stimulated the levator ani muscle at exposures with comparatively little effect on the prostate and seminal vesicles, the standard assay for separating anabolic from androgenic activity in a SARM.
Where the research went
Radius didn’t develop RAD-140 as a muscle drug; its later work was in oncology. Yu et al. (Clin Cancer Res 2017, PMID 28974548) reported AR-dependent growth suppression in AR- and ER-positive breast cancer models, which led to a phase 1 study (LoRusso et al. 2022, PMID 34565686; NCT03088527). Elevated liver enzymes were among the more common adverse events in that trial. Jayaraman et al. (Endocrinology 2014, PMID 24428527) reported neuroprotective effects in cultured neurons and rats. The case-report literature also includes liver injury linked to unregulated products labeled as RAD-140 (Barbara et al. 2020, PMID 33062783). Those products were of unknown composition, which is exactly why lot-level testing matters.
